Vancomycin inhibits D-Alanine D-Alanine
Yes u are right that peniciliin is structural analogue of D-Alanine D-Alanine . Remember that d- alanine d- alanine is just the sequence in peptidoglycan. transpeptidase cross links these peptidoglycan.
Penicillin block the work of the transpeptidase so they cannot cross link peptidoglycan. Vancomycin however bind the d- ala d - ala unit needed to form the peptidoglycan in the cell wall and so bacterial cell wall is not formed.
So for penicillin the peptidoglycanglycan is formed but not cross linked but in vancomycin u don't form peptidoglycan.
