why sickle cell cause defect in the ability to concentrate urine??

Sickling in the vasa recta interferes with countercurrent exchange in the inner medulla, thereby leading to less free water reabsorption.

Also remember that sickling is enhanced by the normal medullary environment: low oxygen tension, low pH, and high osmolality (which pulls water out of the red cells, thereby increasing the concentration of hemoglobin S).

Sickle cell trait and Sickle cell disease both causes nephrogentic DI. That's why there is low osmolality urine and high dilute urine.
